Chemical Structures and Isolation of Glucosyltransferase Inhibitor from the Leaves of Korean Persimmon

Abstract

As a series of studies to find an anti-plaque substance, three compounds have been isolated from the leaves of Korean persimmons. The studies showed a positive reaction in $anisaldehyde-H_2SO_4\;and\;FeCl_3$ solution, which were confirmed through thin layer chromatography with condensed tannin. The components were identified as (+)-gallocatechin, $gallocatechin(4\alpha\longrightarrow8)-catechin$ and prodelphinidin B-3 by spectroscopic analysis. Their inhibitory effects on glucosyltransferase were also investigated and dimeric prodelphinidin B-3, which possessed a 3,4,5-trihydroxylphenyl moiety in ring B of the flavonoid carbon skeleton, showed a higher inhibitory activity than monomeric (+)-gallocatechin. The inhibitory mechanism of (+)-gallocatechin and prodelphinidin B-3 on glucan synthesis by glucosyltransferase was noncompetitive.
