Abstract
Phellodendri Cortex is a medicinal stuff obtained by drying the bark of Phellodendron amurense RUPR and Phellodendron chinese SCHNEID which belong to rutacease. This study identified antineoplastic activity and immune enhancement activity of extracts of Phellodendri Cortex and their fractions, and proved experimentally that they are useful in antineoplastic and immune activator,as follows. 1. B lymphocyte stimulus activity of 2mg fraction of Phellodendri Cortex was tested through the formation of polyclonal antibody in a C57BL/6XC3H mouse. The result of the test showed that the fraction Fr-2 had the strongest activity, and a 10-20mg of dose was the most effective. 2. The administration of 2mg/100g Fr-2 into a mouse in which sarcoma 180 solid tumor was implanted showed tumor inhibition rate of 95.3% which tended to be dependent on concentration. 3. B lymphocyte stimulus activity of the fraction Fr-2-3-2 apleared to be very excellent. In the composition, carbohydrate accounted for 100%. 20mg administration brought out maximum stimulus activity. 4. In the review of chemical luminescence activity of phagocytes by concentrations of the fractions Fr-2, Fr-2-3 and Fr-2-3-2, the chemical luminescence increased from PEC on the fifth day, eighth day and ninth day, respectively, after feeding 20mg to each mouse through mouth. The activity was maximum on the eighth day. It began increasing on the second day and continued increasing until the eleventh day. It was found that the formation of phagocvtes in spleen was dependent on dosage. 5. Then, the effect on the cell toxicity and leady enzyme of blood of the fractions Fr-2, Fr-2-3 and Fr-2-3-2 was examined. It was found that endoperitineal administration of 950mg/kg extract had no effect on S-GOT, S-GPT and S-ALP twenty-four hours later, and there was no effect on the toxicity and leaky enzyme of blood of Fr-2,Fr-2-3 and Fr-2-3-2. In conclusion, this study identified B lymphocyte stimulus activity, phagocyte activity and antineoplastic activity of Phellodendri Cortex which are found to be dependent on concentration.
