Identification and Characterization of Flavanone 3-Hydroxylase (F3H)Gene from Dendranthema grandiflora

Abstract

A cDNA clone for a flavanone 3-hydroxylase (F3H) gene was isolated from Dendranthema grandiflora. The cloned F3H gene was 1,262 bp long encoding 357 amino acids. The deduced nucleic acid sequence showed 73% identity to F3H gene of Arabidopsis thaliana. The reverse transcriptase polymerase chain reaction (RT-PCR) showed that the F3H gene was expressed in yellow petals of Dendranthema grandiflora while it was not expressed in white petals of Dendranthema grandiflora. Low stringency of DNA blot analysis indicated that there was a single copy of gene encoding F3H in Dendranthema grandiflora.

Flavanone 3-Hydroxylase를 위한 cDNA clone은 국화로 부터 분리되었다. Clone된 F3H 유전인자는 1,262bp이고, 357개의 아미노산으로 구성되어 있다. 밝혀진 nucleic acid 서열은 Arabidopsis thaliana의 F3H 유전인자의 서열과 73%의 동일성을 보여준다. RT-PCR에서는 F3H 유전인자가 국화의 노란 꽃잎에서는 발현되고, 국화의 흰 꽃잎에서는 발현이 되지 않는다는 것을 보여주고 있다. DNA blot 분석은 국화에서의 F3H유전인자를 encoding하는 유전인자는 single copy이다.
