Effect of Salviae Multiorrhizaei Radix on Cultured Mouse Myocardial Cells Damaged by Hydrogen Peroxide

Abstract

Objectives : To clarify the cardotoxicity and the protective effect of Salviae Multiorrhizaei Radix(SMR) on oxygen free radicals in cultured myocardial cells of neonatal mouse. Methods : Cardiotoxicity was measured by MTT assay after myocardial cells were grown for 5 hours in the media containing 1-30 $\mu$M concentrations of $H_2O_2$. In addition. the protective effect of Salviae Multiorrhizaei Radix(SMR) was determined in these cultures. Results : Cell viability was significantly decreased in dose- and time-dependent manners after hours. In the protective effect, SMR exposure of 15 $\mu$M $H_2O_2$ to cultured rat myocardial cells for 5 prevented the $H_2O_2$-induced cytotoxicity in these cultures. Conclusion : From the above results, it suggests that $H_2O_2$ has toxic effect on cultured mouse myocardial cells and SMR blocked the cardiotoxicity induced by $H_2O_2$.
