Abstract
Purpose : This study was designed to demonstrate the effect of Ojeok-san on annihilation of human uterine myomal cells and cell apoptosis. Method : Uterine myomal cells were collected from a patients with myoma of the uterus. The cells were cultured in a medium containing various different concentrations of Ojeok-san according to time periods and were measured for viability compared to control group(10$\%$ FBS, 0$\%$ FBS). In addition, we conducted the Western blot analysis of caspase-3 and measured the change of a cycle of cell by flow cytometry. Result : 1. As a result of culturing the uterine myomal cells according to $200{\mu}g/ml,\;500{\mu}g/ml,\;1000{\mu}g/ml,\;2000{\mu}g/ml,\;3000{\mu}g/ml$ concentration of Ojeok-san, the number of dead cells increased in proportion to the concentration of Ojeok-san. 2. As a result of culturing the uterine myomaI cells according to 24, 48, 72hr time periods, the annihilation of uterine myomal cells was promoted in proportion to exposure time. For this fact, we took a photomicrograph and verified on the sense of sight. 3. As a result of the Western blot analysis of caspase-3, activation of caspase-3 increased in proportion to time. 4. As a result of the investigation of a cycle of cell by flow cytometry for the effect of Ojeck-san on cell apoptosis, the percentage of sub G1 cells with apoptotic content increased in proportion to time periods. Conclusion : These results suggest that Ojeok-san has the ability to annihilate uteine myomal cell and to promote cell apoptosis. Therefore, it appears that Ojeok-san is appropriate for the treatment of human uterine myomal patients.
