Obtaining of Lily Symptomless Virus-Free Stock by Callus Culture in Lilium Oriental Hybrids

캘러스 배양에 의한 오리엔탈 나리의 LSV 무병주 획득

Abstract

This study was performed to obtain virus-free stock by the callus culture in Lilium oriental hybrids. LSV-free clones of 70.974.2% in Acapulco and 50.063.3% in Casa Blanca were obtained by scale culture and the same clones of 9090.3% in Acapulco and 86.7100% in Casa Blanca were obtained by shoot apex culture with lily symptomless virus (LSV)-infected plants. In plantlets regenerated from callus, during the 1st subculture of callus, LSV-free clones of 60% in Acapulco and 6070% in Casa Blanca were obtained and in the 2nd subculture, LSV-free clones of 100% in both cultivars were obtained. Chromosome number of plants regenerated from callus subcultured for 2 years in Acapulco and Casa Blanca was 2n=24, as the number of mother plants, and those plants in both cultivars were normal in phenotype such as flower shapes and colors, and plant types. The result of this study is useful to production of virus-free plants of Lilium oriental hybrids.

본 연구는 캘러스 배양을 통한 바이러스 무병묘 획득을 목적으로 LSV에 감염된 오리엔탈 나리 아카풀코와 카사블랑카를 식물재료로 사용하였다. 인편 배양과 경정 배양에 의해서는 아카풀코가 70.9-74.2%와 90-90.3%, 카사블랑카0.0-63.3%와 86.7-100% 제거되었다. 캘러스 배양의 경우, 계대 배양 1회 째의 캘러스로부터 재생된 식물체에서는 아카풀코가 65%, 카사블랑카0%, 계대 배양 2회 째의 캘러스로부터 재생된 식물체에서는 두 품종 모두 100% 제거되었다. 또한 2년간의 캘러스 배양 후 재생된 식물체의 염색체 수는 아카풀코와 카사블랑카 모주의 염색체 수와 동일한 2n=24이었으며, 꽃의 형태 및 색깔 그리고 식물체 형태 등 표현형에 있어서도 정상이었다. 본 연구의 결과는 오리엔탈 나리의 바이러스 무병주 생산에 유용하게 활용될 것이다.
