Genetic Relationship Analysis of Melons (Cucumis melo) Germplasm by RAPD Method

Abstract

RAPD analysis was used to determine the genetic relationship of 50 melon (Cucumis melo L.) accessions. Thirty-five random primers (Operon Co.) were tested and 20 of which showed polymorphism were selected. The amplified fragment ranged from 0.25 to 3.00 kb in size. The sum of polymorphic bands scored were 142 (51%), and an average number of amplified bands per primer was 7.0. From a cluster analysis, 50 accessions were separated into two major groups: one was C. melo species (C. melo var cantalupensis, inodorus, reticulatus, flexuosus, agrestis, momordica, chito, dudaim, conomon) and the other was different species (C. myriocarpus, C. prophetarun, C. anguria, C. africanus). Forty six out of 50 accessions that belonged to C. melo group were divided into seven minor groups. Most of the cultivated western melons clustered together consisted of C. melo var reticulatus, inodorus, cantalupensis, and flexuosus. The Korean native melons, which belong to conomon group was clearly different from western group. Four species, C. melo var reticulatus, cantalupensis, inodorus, and flexuosus which belong to western group were difficult to separate each other.

RAPD 마커를 이용하여 이종, 야생종, 재래종, 육성 계통, 재배종들로 이루어진 50개 멜론 유전 자원에 대해 유연 관계를 분석하였다. RAPD 분석에 사용된 20개 primer에서 나타난 DNA 단편 크기는 0.253.00kb 범위에 위치하였다. 총 밴드 수는 278개였으며 이중 다형성을 나타내는 밴드는 142개(51%)였다. 유집 분석 결과 크게 C. melo 종과 이종 그룹으로 분류되었으며, C. melo 종은 다시 cantalupensis, reticulatus, inodorus, flexuosus계, chito/dudaim계, momordica계, conomon계 등으로 구분되었으나, cantalupensis계, reticulatus계, inodorus계, flexuosus계는 한 그룹으로 되어 구분이 곤란하였고, 동양계인 국내 재래종들이 속하는 conomon계는 서양계인 cantalupensis 또는 inodorus 등과는 확연히 구분되었다. 또 flexuosus계는 cantalupensis와 reticulatus 등과 같은 그룹에 속하나 그룹 내의 소그룹 속에 분산되어 있었고, chito/dudaim계라고 알려진 3계통도 변종의 두 그룹으로 나타났다.
