Effect of Hwangryunhaedok-tang and its modified preseription on the recovery of spatial cognitive function in the brain ischemia induced by four-vessel occlusion in rats

Abstract

Objectives : The purpose of the study we report here was to determine recovery of spatial cognitive of HwangRyunHaeDok-Tang (Huang-lian-jie-du-tang, HT, 黃連解毒湯) and its modified prescription(HTM) on global ischemia induced by 4-vessel occlusion in wistar rats. Method : The sample of HT and HTM were prepared by lyophilization after extraction with $100\%$ water. Rats were induced to 10 minutes of forebrain ischemia by 4-vessel occlusion (4-VO) and reperfused again. They were administered at a dose of 300mg/kg to 0 minutes and 90 minutes after 4-VO. Functional improvement was evaluated with the Morris water maze at 7 days after ischemia. Rats were sacrificed after behavior test and the number of CAl pyramidal neurons were counted in hippocampal sections stained with cresyl violet. Result: Compared to control group, HT and HTM groups confimed that contributes to behavioral improvement. The administration of HT and HTM showed a significant neuroprotective effect on hippocampal CAl neurons after ischemia compared to cotrol group. Conclusion : The results of this study demonstrate that administration of HT and HTM is effective in reducing neuronal damage and functional recovery in response to transient forebrain ischemia.
