Study on the Antibacterial Effects of Silver Nanoparticles Prepared by Sonication Method

Abstract

The antibiological effect of nanoparticles on staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) using silver nanoparticles (Ag-NPs), < 10 nm, prepared by the sonication of $AgClO_4$ was studied. To determine the antibacterial activity, we applied $100\;{\mu}L$ of 0.054% Ag-NPs solution to each culture medium containing test organism and evaluated the antibacterial activities by the cup diffusion method. Only Ag-NPs stabilized with poly(N-vinyl 2-pyrrolidone) [PVP] showed antibacterial activity and the minimum inhibitory concentration (MIC) to S. auresu and E. coli were 5 ppm. And the complete sterilizing concentrations for the test organisms were 50 and 100 ppm, respectively. Also we observed the morphological alteration and cell damages caused by Ag-NPs treatment using a scanning electron microscopy (SEM). Above results supports the evidence that Ag-NPs have strong antibacterial activity to S. aureus and E. coli.

본 연구에서는 염화은($AgClO_4$)을 초음파 처리하여 얻은 10 nm 이하의 은 나노입자의 항균력을 측정하기 위해 그램 양성균인 Staphylococcus aureus (S. aureus)와 그램 음성균인 Escherichia coli (E. coli)를 배양한 배지에 0.054%은 나노용액 $100\;{\mu}L$를 넣고 cup diffusion method로 실험을 행하였다. 그 결과 poly(N-vinyl 2-pyrrolidone) [PVP]로 안정화시킨 은 나노 입자에서만 항균활성을 보였으며 S. aureus와 E. coli에 대한 최소 억제농도(minimum inhibitory concentration, MIC)는 5 ppm이었으며 완전 살균농도는 50 ppm과 100 ppm이었다. 은 나노입자에 의해 사멸된 균의 형태상 변화와 세포 손상을 scanning electron microscopy (SEM)으로 확인하였다. 또한 은 나노 입자가 양성균(황색포도상 구균)과 음성균(대장균)에서의 특수한 향균 효과를 증명할 수 있었다.
