Hydrogen Peroxide Induces Resistance against Anthracnose in Cucumber (Cucumis sativus)

Abstract

By using a native polyacrylamide gel electrophoresis and determining the enzyme activity of intercellular wash fluid protein extracted from cucumber leaves, it was demonstrated that 50 $mmoláL^{-1}$ of hydrogen peroxide treatment resulted in the accumulation and activation of peroxidase and chitinase. Through the challenge inoculation with Collectotrichum orbiculare, it was also confirmed that hydrogen peroxie suppressed the anthracnose development on cucumber leaves. The increase in resistance against cucumber anthracnose could be the result of increased peroxidase and chitinase activity because both enzymes are known for helping plants resist disease. The patterns of isozyme accumulation and of systemic activation were, however, different according the enzymes, indicating that hydrogen peroxide activates differently peroxidase and chitinase. It was suggested that hydrogen peroxide may have a direct influence on peroxidase activity, but an indirect influence on chitinase activity via a signal molecule synthesis which may activate chitinase.
