Identification of Deformed Wing Virus from the Honeybee in Korea and Establishment of PCR Detection Method

Abstract

Deformed wing virus (DWV) causes a wing deformity and a premature death of an adult honeybee. Although those symptoms were found in korean honeybees, this is first report, in our knowledge, that the infection the DWV have been presented in Korea. Rapid detection system may be needed for the management of hive because DWV generally persists as a latent infection with no apparent symptom and there is no available control method for this disease yet. In this study, our laboratory confirmed the presence of the DWV using PCR detection method from adult honeybees which have wing deformities. PCR product size is 479 base pairs with a specific primer set(DWV-SF/DWV-SR). With T-vector cloning, the sequence of the PCR product was analyzed. And then, Real-Time PCR detection method for a rapid detection of DWV was developed. The PCR system was able to detect easily the existence of the virus from honeybee sample. Specific PCR method showed improved sensitivity in the artificially contaminated templates.
