The expression and interaction of HLA-G and IL-10 in cervical cancer

자궁경부암에서 HLA-G와 IL-10의 발현 및 상호작용

Abstract

=목적: 본 연구의 목적은 자궁경부암 환자의 조직에서 HLA-G 유전자 및 IL-10 유전자의 발현을 reverse transcriptase- polymerase chain reaction (RT-PCR) 및 western blot analysis를 통해 알아보고, HLA-G와 IL-10 발현의 상관성과 자궁경부암의 임상병리학적 예후인자와의 연관성을 알아보는 것이다.연구 방법: 2004년 10월부터 2005년 10월까지 자궁경부암으로 진단받은 40명의 환자와 정상환자 15명의 자궁경부 조직을 대상으로 하였다. HLA-G의 mRNA의 측정은 quantitative real time RT-PCR을 이용하였고, IL-10 mRNA의 경우에는 semi-quantitative RT-PCR을 이용하였다. 그리고 HLA-G 및 IL-10의 단백질 발현은 western blot analysis를 통해 측정하였다. 결과: HLA-G 및 IL-10의 mRNA 발현은 정상 자궁경부 조직에 비해 자궁경부암 환자의 조직에서 통계학적으로 유의하게 더 높게 나타났다 (P<0.001, P<0.001). HLA-G와 IL-10의 단백질 발현 또한 자궁경부암 조직에서 더 높은 발현을 보였다 (P<0.001, P=0.041). HLA-G의 mRNA 발현은 IL-10의 mRNA 발현과의 상관성은 통계학적으로 유의하지 않았지만, 상관성 경향을 보였다 (P=0.061). 자궁경부암의 임상병리학적 예후인자와의 연관성에서 HLA-G의 mRNA 발현은 FIGO 병기와 연관성이 있었으며, 초기 병기에서 HLA-G mRNA 발현 수치가 더 높게 나타났다 (P=0.045)
