Antioxidant and Antimicrobial Capacity of Several Monofloral Honey Correlation with Phenolic and Flavonoid Contents

Abstract

This study aimed to evaluate the antioxidant and antibacterial activity of several monofloral honeys which were harvested in South Korea. First at all, we determined important chemical parameters such as pH, moisture, HMF and Sugar. Honey samples also were analyzed for total phenolic contents through a spectrophotometric determination with a modified Folin-Ciocalteu method and total flavonoid content determined with aluminium chloride. Total phenolic compound was highest in Fagopyrum esculentum honey (171$\mu$g GAE/g) and flavonoid content was the highest in Brassica campestris honey (102$\mu$g QUE/g) than other samples. The antioxidant activity was evaluated by FRAP (ferric reducing-antioxidant assay) test. The FRAP is based on the reduction of ferric 2,4,6-tris (2-pyridyl)-1,3,5-triazine [Fe(III)-TPTZ] by spectrophotometric analysis. Fagopyrum esculentum honey had the highest antioxidant activity (27$\mu$M mM Fe(II)/g) while Robinia pseudoacacia honey had the lowest antioxidant activity (4.5mM Fe(II)/g). The antimicrobial activity obtained for the spectrophotometric method MIC value against Gram-positive and Gram-negative bacteria. The results showed that dark honeys present a highest antimicrobial activity while the clear honeys showed a lowest antimicrobial activity.

국내에서 생산된 밀원별 벌꿀의 항균 및 항산화활성을 조사하여 기능성 자연식품으로서의 벌꿀의 효능을 조명하고자 이 연구를 수행하였다. 먼저 밀원별 벌꿀의 pH, 수분, HMF, 자당 분석을 통해 밀원별 벌꿀의 기본적인 특성을 이해하였으며, Folin-Ciocalteu방법에 따른 총 페놀 함량과 aluminium chloride를 통한 플라보노이드 함량 분석을 통해 항산화 활성과의 상관관계를 알아보았다. 그 결과 총 페놀화합물의 함량은 메밀(Fagopyrum esculentum)꿀이 가장 높았으며(171$\mu$g GAE/g), 총 플라보노이드 함량은 유채(Brassica campestris)꿀이 가장 높은 것으로 나타났다(102$\mu$g QUE/g). 이들 시료에 대한 항산화활성은 FRAP (ferric reducing-antioxidant assay) 활성을 통해 전자공여능력 측정함으로써 확인하였다. 그 결과 메밀꿀이 27$\mu$M mM Fe(II)/g로 가장 높은 것으로 나타난 반면 아카시아(Robinia pseudoacacia) 꿀이 4.5mM Fe(II)/g로 가장 낮은 것으로 확인되었다. 밀원별 벌꿀 시료에 대한 항산화 활성은 페놀 및 플라보노이드 함량과 깊은 상관관계를 보이고 있었으며, 총 단백질 함량과도 높은 상관관계를 보이고 있었다. 밀원별 벌꿀에 대한 항균활성은 그람 음성 및 양성 균주에 대한 MIC test를 실시하였다. 그 결과 메밀꿀의 항균활성이 가장 높은 것으로 나타났으며, 특히 그람 양성균인 Bacillus subtilis 균주에서 가장 높은 항균활성을 보이는 것을 확인할 수 있었다.
