Variation of Glucosinolate Content in the Root of Susceptible and Resistant Chinese Cabbage Cultivars during Development of Clubroot Disease

Abstract

Variations of major glucosinolate (GS) contents in root among 35 clubroot-resistant and -susceptible Chinese cabbage (Brassica campestris ssp. pekinensis) cultivars cultivated on infected soil were evaluated. Two indolic (glucobrassicin and 4-methoxy-glucobrassicin) and one phenyl GS (gluconasturtiin) were identified as major GSs in Chinese cabbage root. Among the three identified GSs, the contents of glucobrassicin and gluconasturtiin were found to be positively correlated with the clubroot disease severity ($r^2$=0.519), while the 4-methoxy-glucobrassicin content was negatively correlated ($r^2$=0.684). After artificial infection with P. brassicae, the glucobrassicin and gluconasturtiin contents in root increased after 20 days in only the two susceptible cultivars 'Chilsung' and 'Kangryeok-Yeorum', while the content changes in the two resistant cultivars 'CR-kangta' and 'CR-saerona' were not significant. The 4-methoxy-glucobrassicin content significantly decreased in the two susceptible cultivars from 20 days after infection, showing the inverse fluctuation compared to the other GSs. Consequently, we could detect the characteristic positive or negative correlation of individual GS with clubroot disease severity and rapid fluctuation of GS contents after infection in Chinese cabbage root.

배추 뿌리혹병 저항성과 감수성 35품종을 뿌리혹병 이병 포장에 재배하여 각 품종별 발병도와 뿌리 내 indolic 및 phenyl glucosinolate 함량 변이를 조사하였다. 또한 저항성('CR-강타', 'CR-새로나')과 감수성('칠성', '강력여름') 각 2품종에 뿌리혹병 균을 인위적으로 접종한 후 체내 glucosinolate 함량변화를 접종하지 않은 건전 식물체와 비교하였다. 배추 뿌리에서는 2종의 indolic 및 1종의 phenyl glucosinolate가 동정되었다. 동정된 3종의 glucosinolate 중 glucobrassicin과 gluconasturtiin은 뿌리혹 발병도와 유의적인 정의 상관관계를 보였으며($r^2$=0.519), 4-methoxy-glucobrassicin 함량은 부의 상관을 보였다($r^2$=0.684). 뿌리혹병의 인위적 접종 20일 후, 뿌리내 glucobrassicin과 gluconasturtiin 함량은 감수성인 '칠성'과 '강력여름'에서 유의적으로 증가한 반면 저항성 품종들에서는 함량의 차이가 없었다. 반면, 4-methoxy-glucobrassicin 함량은 접종 20일 후 감수성 품종에서 오히려 감소하였으며 저항성 품종에서 증가하는 경향을 보여, 타 두 물질과 상반되는 특이한 변화양상을 보였다. 결과적으로, 본 연구를 통해 배추 뿌리 내에 동정된 3종의 glucosinolate는 발병도와 정의 또는 부의 상관관계를 보였으며 접종 후 경시적인 변화 역시 이와 동일한 양상을 나타내는 것을 확인하였다. 따라서 동정된 이들 3종의 glucosinolate는 뿌리혹의 발달과 밀접하게 연관되어 있으며 저항성과 감수성을 판별할 수 있는 지표로서 이용 가능한 것으로 판단된다.
