Antioxidant and Antigenotoxic Effect of Sansuyu Fruit (Corni fructus) Extracted with Various Solvents

Abstract

The objective of this study was to evaluate the antioxidant activities and antigenotoxic effect of Corni fructus extracted with various solvents (Ace; acetone, EtOH: ethanol, and MeOH; methanol). Total phenolic content (TPC), DPPH radical-scavenging activity (RSA), and superoxide dismutase (SOD)-like activity were determined in the samples. Also antigenotoxic effect of Corni fructus was determined by measuring inhibitory effect of $H_2O_2$ induced DNA damage using comet assay in human leukocytes. The TPC of the Corni fructus extracts was in order of Ace (0.5 g GAE/100 g) $IC_{50}$ of DPPH RSA was significantly different between EtOH, MeOH and Ace, especially Ace (871.1 ${\mu}g/ml$) is two-fold lower than EtOH (458.9 ${\mu}g/ml$) and MeOH (468.2 ${\mu}g/ml$), and the IC50 of SOD-like activity showed similar reduction (EtOH; 827.2 ${\mu}g/ml$=MeOH; 1193.8 ${\mu}g/ml$ ${\mu}g/ml$). MeOH extract of Corni fructus showed the highest ORAC activity among samples. $200{\mu}M$ $H_2O_2$ induced DNA damages in human leukocytes were significantly reduced by the pretreatment with all Corni fructus extracts. Significantly high correlation coefficients were observed between TPC and antioxidant paramaters, but $H_2O_2$ induced DNA damage did not have a relationship with TPC and antioxidant paramaters. As a results, the Corni fructus extracts have antioxidant activities and antigenotoxic effect and its activities showed that there were difference between solvent extractions. Further inverstigation is needed to identify its antigenotoxic compunds.
